    The goalThe course provides a strong background to enzyme technology and contemporary techniques including enzyme kinetics and modelling of enzymatic reactions, large-scale enzyme production and purification, the application of recombinant DNA technology in enzyme production, procedures for obtaining novel industrial enzymes, immobilization of enzymes, as well as basic principles for optimization and design of enzymatic processes.
    The outcomeAfter the course, the students will gain knowledge of basic principles for optimization, modeling and design of bioprocesses in which both, free and immobilized enzymes play a role. By mastering enzyme purification and other techniques for enzyme assay and recovery as well as the numerical methods to analyze enzyme kinetic data with the advance of a computer, the students will have a strong background in different practical skills to formulate and solve problems in the field of enzyme technology.
    Contents of lectures• An introduction in Enzyme technology and basic principles of biocatalysis. • Enzyme production (Enzyme sources: animal and plant enzymes; microbial enzymes; Biochemical fundamentals; Factors affecting biosynthesis of enzymes; Regulation mechanisms of enzyme biosynthesis; Optimization and technology of industrial enzyme production). • Enzyme isolation, purification and characterization (General principles; methods for cell disruption; removal of whole cell and cell debris; Concentration and primary purification; Column chromatography; Evidence of purity; Scale-up of bulk enzymes production and therapeutic enzymes production; Legislative and safety aspects). • Optimization of industrial enzymes by molecular engineering. • Immobilization of enzymes (Methods and supports; Effect of immobilization on enzyme properties and stability). • Design and modelling of enzyme processes (Limitations of classical enzyme kinetic models; Performance criteria for the process; Process windows; Rules for choice of reaction parameters and systems; Chemical control of enzyme processes; Enzyme kinetics in homogeneous and heterogeneous systems with calculus). • Industrial enzymes: general characteristics and applications (oxidoreductase, amylases; pectinase, cellulases and hemicellulase, proteases, lipases; aminoacylase; penicillin acylase). • Specific enzymes and their applications in medicine, analytics and others.
    Contents of exercisesCalculus for modelling of enzyme processes. The Lab is designed to provide the students with the necessary techniques for isolation, concentration and primary purification of enzyme such as cell rupture by sonication, removal of cell debris by centrifugation, ultrafiltration, precipitation (solvent, salt, adjustment of solution pH). Enzyme purification in two-phase systems and column chromatography, and enzyme immobilization by several techniques are also included.

    Methods of teachingTeaching methodology consists of lectures with pronounced interactive character, tutorials, probleme solving and demonstrations. Lecturing will be accompanied by VIDEO BEAM presentation. The course is accompanied by coordinated lab exercises, including measurement, tutorials collection, analysis, interpretation and presentation of data.
